How to Reconstitute Peptides

What Is Reconstitution?

 

Reconstitution is the process of dissolving freeze-dried (lyophilized) peptide powder into a liquid solution so it can be accurately measured and administered.

Research peptides are shipped as dry powder because peptides are far more stable in this state. Once dissolved in liquid, the solution degrades over time and must be refrigerated and used within a defined window.

 

Supplies You’ll Need

 

  • Peptide Vial: Sealed glass vial containing dry powder with label indicating total mass (e.g., BPC-157 5mg).

  • Bacteriostatic Water (BAC Water): Sterile water with 0.9% benzyl alcohol preservative to inhibit bacterial growth over 25–30 days.

  • Insulin Syringes: Standard U-100 syringes (1.0 mL for adding water; 0.5 mL or 0.3 mL for daily micro-dosing).

  • Alcohol Swabs: 70% isopropyl alcohol prep pads for mandatory surface sterility.

  • Sharps Container: Puncture-resistant container for safe needle disposal.

How Much BAC Water to Add

 

The volume of BAC water added determines the liquid concentration.

 

The Formula

 

Concentration (mg/mL) = Total Peptide in Vial (mg) / BAC Water Volume Added (mL)

 

Standard Volumes

 

  • 1.0 mL: Creates a highly concentrated solution. Ideal for small-mass vials (1–3 mg) with micro-gram dosing.

  • 2.0 mL: The most common standard. Balances precision with reasonable volume for 5 mg and 10 mg vials.

  • 3.0 mL to 5.0 mL: Used for high-mass vials (20 mg+) or when higher dilution is required for precise tiny draws.

Step-by-Step Reconstitution Process

 

  1. Prepare Workspace: Clean your surface and wash hands thoroughly. Ensure the vial is at room temperature.

  2. Sterilize Stoppers: Wipe the rubber stoppers of both the BAC water and peptide vials with alcohol swabs and let air dry.

  3. Draw BAC Water: Using a fresh syringe, draw the planned amount of BAC water (e.g., 2.0 mL = two 100-unit draws). Clear any large air bubbles.

  4. Inject BAC Water: Insert needle through the stopper at a slight angle. Aim the liquid stream against the inner glass wall—never squirt directly onto the powder.

  5. Dissolve Gently: Remove syringe. Swirl gently or let sit for 1–3 minutes. Never shake the vial.

  6. Inspect Solution: Check against light. The liquid should be completely clear and particle-free.

  7. Label & Refrigerate: Mark the vial with reconstitution date and water volume. Refrigerate immediately.

Drawing a Dose from the Reconstituted Vial

 

  1. Wipe the rubber stopper with an alcohol prep pad.

  2. Draw air into a fresh syringe equal to your target dose volume.

  3. Inject the air into the upright vial to equalize internal pressure.

  4. Invert the vial, submerge the needle tip in liquid, and draw slightly past your target line.

  5. Tap the barrel to move air bubbles to top, push excess air back into vial, and line up plunger to exact draw mark.

Concentration Math: Worked Examples

Example 1: BPC-157 (5 mg Vial)

 

  • BAC Water Added: 2.0 mL

  • Concentration: 5 mg / 2.0 mL = 2.5 mg/mL

  • Target Dose: 250 mcg (0.25 mg)

  • Dose Volume: 0.25 mg / 2.5 mg/mL = 0.10 mL

  • Syringe Draw Mark: 10 Units on a U-100 syringe

Example 2: Semaglutide (3 mg Vial)

 

  • BAC Water Added: 3.0 mL

  • Concentration: 3 mg / 3.0 mL = 1.0 mg/mL

Special Cases

 

  • Blend Vials: Reconstitute normally. Math reflects combined total mass; ratio of compounds remains fixed.

  • IU-Dosed Compounds: Calculate concentration as IU per mL rather than mg per mL.

  • Small Vials (1–2 mg): Use lower diluent volume (0.5–1.0 mL) so draw volumes aren’t too small to visually measure.

Common Mistakes to Avoid

 

  • Shaking the Vial: Shearing forces can degrade delicate peptide structures and create excess foam.

  • Direct High-Pressure Jetting: Squirting water forcefully directly onto the powder can damage proteins.

  • Forgetting to Equalize Pressure: Failing to inject air first creates a vacuum inside the vial, making dose draws inaccurate.

  • Reusing Syringes: Dulls needles and introduces contamination into multi-dose vials.

  • Room-Temperature Storage: Unrefrigerated liquid solutions degrade rapidly.

Troubleshooting

 

  • Powder Won’t Dissolve: Wait 10–15 minutes and gently roll the vial between hands. Do not force or shake.

  • Cloudy or Colored Solution: If solution remains milky, discolored, or contains floating specks after 15 minutes, discard the vial due to potential contamination or breakdown.

  • Wrong Water Volume Added: Do not panic—simply recalculate concentration based on the actual liquid volume added.

  • Foam Layer: Allow vial to rest upright in the refrigerator for 20 minutes until foam settles. Draw liquid from beneath the foam line.

Post-Reconstitution Storage

 

  • Temperature: Keep strictly at 2–8°C (36–46°F).

  • Shelf Life: Usable for 25–30 days when mixed with BAC water.

  • Position: Store upright to prevent compound contact with the rubber stopper.

  • Never Freeze: Do not freeze liquid solutions. Ice crystals will break molecular bonds.
